Abstract
Antibody therapeutics targeting amyloid-beta (Aβ), such as Aducanumab and Lecanemab, are among the first Alzheimer's disease treatments to show clinical potential, with efficacy thought to depend partly on Fc receptor-mediated immune clearance of Aβ plaques. Testing new antibody variants in mouse models is slow and expensive, so TREAT-AD's ADHTS core develops cell-based assays to screen candidates earlier. This project lays the groundwork for a scalable, cell-based high-throughput screening assay of antibody-mediated Aβ clearance. We purified Fc-competent and Fc-silenced Aducanumab, confirmed its specificity for full-length Aβ, and established a live-imaging assay of Aβ uptake in microglia. Assay optimization is ongoing.
